The genomes of higher organisms encode thousands of long noncoding RNAs (lncRNAs); however, the overwhelming majority of these RNAs are uncharacterized – their functions are a mystery. Jeffrey’s work aims to better understand how lncRNAs accomplish their diverse cellular roles, focusing on the roX lncRNAs in particular. The roX RNAs are essential to regulating gene expression from the X-chromosome in male fruit flies. Using biochemical, structural, genetic, and next-generation sequencing technologies, he plans to build a mechanistic map of how roX and other lncRNAs function.
